San Marco cathedral - it has the most magnificent golden and enamelled retable (altar screen) you ever saw - known as the Pala d'Oro, most of it was looted from Byzantium by the 4th Crusade in 1204 - it's behind the altar. The Bronze horses also looted in 1204 are in the San Marco museum - the ones out the front are replicas.
San Marco itself is definitely worth seeing - it's eleventh century, Byzantine style, covered in mosaics.
Haven't been to Venice myself, I envy you.
